37度放心切，没问题。不要小看BamHI，这个酶的效率很高的。一般有星活性的酶都不担心切不开，而是担心切多了=.=如果一定担心温度的问题，或者最后实验结果是37度切不开，则考虑以下两种方法：1. BamHI和XhoI都是常用酶，比较便宜，可以换一家公司买。至少Fermentas和NEB的BamHI都是37度的。2. 分布酶切。单酶切，跑胶回收，再另一个单酶切。这要求你的DNA template多一点，比如质粒或者PCR产物。
