番木瓜通常采用实生繁殖，因而在育苗过程 中出现大量的“木瓜公”，即雄株，植株只有在6― 8个月后开花时才能鉴别性别，在此之前并不能 从形态上准确判断，通常情况下雄株个体约占 3％～5％，必需在开花时拔掉，这种情况不仅对生 产造成极大的浪费，而且在有“木瓜公”的果园留 种会使以后繁殖出的幼苗出现大量的“木瓜公”， 危害甚大，因此研究番木瓜的性别决定及其鉴定 意义重大。本文就此方面的新进展进行综述。 　　 　　1、番木瓜花性的多型性 　　 番木瓜基本花型可分为雄花、雌花和两性花 3种，因此番木瓜的株型也就相应分为雄株(male 或staminate)、雌株(female或pistillate)和两性株 (hermaphrodite)三种基本类型，栽培品种主要分 为两性株及雌性株两种类型，目前我国主栽的“穗 中红”品种即属于两性株．两性株栽培广泛，但它 的实生后代性别经常发生分离，分离比一般为两 性株：雌株2：1，植株只有在生长6-8个月后开 花时才能判断性别。 除了常见的3种基本花型外，番木瓜尚有其 他花型，呈现出十分广泛的多型性，并且也有“木 瓜公”结果、两性株上也有雄花的报道(即雌花、雄 花、两性花同时着生在一株植物上的杂性同株现 象)，由此可见，番木瓜的花型表现及遗传是非常 复杂的。 在自然界，雌雄异株是普遍存在的，如高等 植物中的银杏、猕猴桃、芦笋、罗汉果、红豆杉等 都是雌雄异株植物，大多数雌雄异株植物的性 别决定并不像动物那样在胚胎时期就已决定， 而是在生长、分化和发育成熟后的某个阶段才 能确定，同时还会不同程度地受外界环境条件 如营养、温度、湿度、光强、日照时间和植物激素 等因素的影响，因此许多高等植物的性别分化 具有不稳定性，对于番木瓜而言，其花型、性别 也会因季节、高温、不恰当的修剪而变化，如雄 株会变成雌株或两性株，即是受环境因素影响 的结果。 不同性别的番木瓜植株，其结果性能、果实形 态及品质均存在较大的差异。由于雄株并不能结 果，因此应尽量设法避免“木瓜公”的出现。 　　 　　2、番木瓜性别决定及其鉴定研究 　　2．1 形态鉴定 在苗期鉴定植株的性别，对番木瓜的育种及 栽培均具有一定意义。在幼苗形态上，各种类型 的株型几乎没有区别，只有开花时根据花器形态 加以判断。不同品种性别的番木瓜的结果习 性、果实形态及品质均存在较大的差异，因此采 用形态学鉴定的方法在生产中的应用十分有限， 有人试图用物理、化学及组织培养方法来预测植 株的性别，但效果均不理想。 　　 　　2．2 对于性别遗传规律的研究 伴随着遗传学研究水平的提高，植物的性别 研究开始成为关注的重点，同时植物的性别研究 一直是世界植物研究上的薄弱环节，解读植物性 别遗传信息也一直是研究人员梦寐以求的事情， 对于番木瓜，尽管它是二倍体，只有9对染色体 (2n=18)，基因组也很小，约372 Mbp，但直到 目前，人们对它的遗传学研究和认识还十分肤浅。 对于番木瓜性别遗传规律的认识，早在60年 前已有人提出它的性别是由3个等位基因控制的 假说．根据3种株型杂交后代性别分离的比例， Storey等认为3个等位基因分别是M1、M2 及m，它们分别控制雄性、两性及雌性性状，其中 M1、M2为显性，m为隐性，雄株、两性株及雌株的 基因型分别为M1m、M2m及mm，而基因型为 M1M1，M1M2及M2M2的合子均表现为败育，并定位了性别决定基因。显性组合导致显性败育，从而 使两性株的后代发生2：1的分离。后来Storey 继续修正了该假说，认为性别决定并非单个基因 作用的结果，而是受聚集在性染色体上十分狭窄 范围内的一个基因复合体所控制，Hofmeyr进一 步报道了M1和M2是染色体上丢失了性别决定 关键基因的有轻微长度差别的失活区，根据属内 杂交结果，Horovitz和Jimenez指出番木瓜的性 别决定是XX-XY型，雄性基因型为XY，雌性为 XX，两性为XY2，Y2是突变了的Y染色体。最近， Liu等认为性染色体的演变包括了性别决定基 因附近的染色体区域重组抑制，一个永久杂合的 染色体区域或许积累有害的隐性突变并加以固 定，通过在Y染色体上较近的有利突变选择一段 时间后，这些变化或许导致Y染色体的衰退和与 X染色体的分离，在人类中已发现仅有5％的Y 染色体还显示有X-Y染色体的重新交换。因此 认为番木瓜包含一个原始的Y染色体，其上仅有 的雄性特异区域(约占该染色体长度的10％)也 经历了严格的重组抑制和DNA序列的改变，这一 发现进一步为性染色体起源于常染色体提供了直 接证据。 另外一个假说认为3种等位基因分别编码不 同的反式作用因子从而指导不同的花型形成， 将性别决定由基因水平发展到蛋白质翻译水平， 　　 　　2.3 连锁遗传图的构建 高密度的连锁遗传图不仅是分离克隆目标基 因的第一步，而且也是遗传研究的重要工具，连锁 遗传图的构建对克隆相关基因和分子标记辅助选 择具有意义。目前很多作物的连锁遗传图已构建 完成。 番木瓜连锁遗传图的构建远远落后于其他作 物，第一张遗传图在60年前构建，其中只包括性 型、花色、茎色三个形态标记。1996年Sondurt 报道了第二张基于分子标记的遗传图，在该遗传 连锁图谱中，共有62个RAPD标记，性别决定基因 定位于第一个连锁群，其位点的两边各有一个标 记，遗传距离为7cM，最近夏威夷大学的HaoL等 根据Kapoho和SunUp的54株F2群体构建了第三 张高密度分子连锁遗传图，该图谱共有1 501个 标记，其中包括1498个AFLP标记和番木瓜环斑 病毒外壳蛋白标记、性别的形态学标记和鲜果 颜色标记等。这些标记分为12个连锁群，共覆盖 3 294．2cM的基因组，标记间的平均距离为2．2 cM。对这些标记进一步研究，发现在性别决定位 点附近共有225个标记与性别基因共分离．几乎同 时，Chadesworthf构建了非常详细的携带性别决 定基因的染色体图谱，从图上看出番木瓜的性别 决定基因位于该图中一个非常集中的重组区域 内，与其他的雄性特异区(如哺乳动物的Y染色 体)相比，该区域仍然是番木瓜基因组的一小部 分，其研究结果与Hao相吻合，进一步证明性别 决定基因及其附近的标记是紧密连锁的， 　　 　　2．4 与性别连锁的DNA分子标记研究 分子标记技术作为一种基本的遗传分析手 段，已在动物、植物、人类遗传作图和基因分析等 诸多领域广泛应用，分子标记是以生物大分子，尤 其是以生物体的遗传物质――核酸的多态性为基 础的遗传标记(geneticmarkers)，是鉴别基因组中 基因位点(10CUS)的有效手段，分子标记由于具 有能够稳定遗传，遗传方式简单，可以反映生物个 体和群体特征的特点，因而它是目前发展最为迅 速的一类遗传标记。 　　分子标记辅助选择技术是分子标记技术在育 种实践中的具体应用，已在许多作物的早期选择 上有成功的报道，这也激励着研究人员开发早期 剔除番木瓜雄株的分子标记选择技术。由于番木 瓜性别的多型性和偶尔的性别转换以及性染色体 的突变缺失，故其性别的分子水平研究一直是研 究的热点和难点，现已在性别决定及其鉴定方面 取得了一些进展．夏威夷大学的Deputy等率 先获得了成功，他们获得了与番木瓜性别紧密连 锁的3个RAPD标记，经测序，合成引物，转换为 SCAR标记，并认为SCARTl2和SCARWll在两 性株和雄株之间能够产生特异的PCR产物，而在 雌株却极少出现；SCARTI在所有的株性间均有 该PCR产物产生，因此，利用这3对引物可以很 好地区分3种番木瓜株型。基于此他们成功地开 发了检测番木瓜株型技术，即用T1作为正对照， 用Wll或T12鉴定两性株和雄株，在苗期进行早 期性别鉴定，预测番木瓜的性别，准确率为99．2％， 达到了早期选择的目的，后来Parasnis、Lemos和 Uraski也分别成功地开发了雄性(Male-specific)、 两性(hermaphrodite-specific)和将雌性与雄性、两 性区分的分子标记技术及标记辅助选择技术，并用 于性别的早期选择。Eliana等利用RAPD 技术区分了商品栽培品种Solo的3个性别的后 代，发现引物BC210产生的标记BC210438可以检 测供试材料的两性株类型。随后，SomsriI等应用 DNA扩增指纹技术对番木瓜的性别基因标记也 进行了研究。 　　真核生物基因组中的小卫星和微卫星DNA 具有丰富的多态性，由于它们在整个基因组上分 布广泛、均匀，具有保守性强、共显性遗传、能够提 供大量稳定的遗传信息以及分析方便快速等一系 列优点，因此已经广泛应用于生物遗传作图、群体 遗传学研究等方面。印度的Parasnist3等最近利 用微卫星和小卫星探针来鉴定番木瓜的性别，他 们认为有一个微卫星探针(GATA)4在所研究的 番木瓜品种中表现了性别的特异性，从而开发了 苗期番木瓜性别鉴定的微卫星标记，同时，也发现 番木瓜的X、Y染色体的遗传物质完全不同的原 因是由于性别分化过程的不同所导致的，从染色 体水平上揭示了性别差异的遗传物质基础。 利用分子标记研究番木瓜的性别，我国也有 学者进行了探索，陈中海采用同工酶技术、蛋 白质SDS-PAGE方法研究番木瓜雌株、雄株和两 性株3种株性之间的差异，结果发现雌株、雄株和 两性株在过氧化物酶(POD)、酯酶(EST)和多酚氧 化酶(PPO)同工酶酶谱上存在差异，酶活性总的 趋势是雌株活性较强，雄株和两性株较弱，并且 与雌株和两性株相比，在雄株叶柄中多一条afl。 0．29的过氧化物酶同工酶带，在成熟叶片中多一 条Rf=0．75的酯酶同工酶特异带，少一条Rf= 0．35的多酚氧化酶谱带，这提示该3种特异的谱带 可作为鉴别番木瓜株性的参考依据，周国辉”应 用混合分离群体分析法，在205个10碱基随机 引物中，寻找到了番木瓜两性基因(M2)的RAPD 标记一OPQ071800和OPE061050，用这两个引物 检测了210株番木瓜单株DNA，结果表明， OPQ071800和OPE061050与M2基因紧密连锁， 并且位于M2基因的两侧，其遗传距离分别为4，5 cM和1，9cM。该研究为M2基因的克隆、定向育 种开辟了新道路，也使传统的育种工作跨上新台 阶。 　　 　　2．5 其他方法的性别鉴定 除了分子标记辅助选择技术用于性别鉴定之 外，还有人报道用一些生理生化的方法进行早期 性别鉴定，Awadt3]以三月龄的Fairchild品种叶 片和叶柄为材料，通过分析其化学成分的差异，探 讨了番木瓜叶片的化学成分与性别表现之间的关 系，结果发现，雄株与雌株叶片的叶绿素可溶性 固形物、总酸和总酚在含量上没有显著差异，但可 以通过雌株叶片和叶柄中较高的干物质、硅、吲哚 及叶片氨基酸和游离脯氨酸含量，进行雌性株的 早期鉴定，雄株则可以通过叶片和叶柄的较高的 灰分物质含量，及叶柄中较高的过氧化物酶和游 离脯氨酸含量来预测，还有许多不同的研究人员 从不同的方面又发展了一些其他的方法对番木瓜 的幼苗进行了早期鉴别。 另外，栽培管理上的一些措施也可以影响番 木瓜性别表现，Suranant用NAA和GA-3处理 番木瓜的幼苗，发现适当浓度的NAA处理可以 减少群体中雄株的百分率，GA-3处理虽然没有 改变雄雌株的比例，但雄性特征的出现早于其他 两种性型。因而，可通过适时喷洒化学药剂NAA 和GA―3，达到雄株的早期鉴别和降低雄株率的 目的，Mitra也报道用100μg／gNAA处理幼苗 可以提高雌株的百分率(由46％提高到62，5％)， 支持了上述观点， 　　 　　3 花器的发育 　　 为了弄清番木瓜独特的花器发育过程，有研 究者对番木瓜两性株花器形成过程的形态变化进 行了观察，我国台湾的谢明宪等用解剖显微镜 及扫描电子显微镜观察了台农二号番木瓜两性株 各龄花芽的发育过程．根据形态，将两性花的花 器形成过程分为4个阶段：萼片及花瓣原基相继 出现的花被原基形成期(第1～3节)，雄蕊及心皮 原基相继开始形成的雌雄花器原基初形成期(第 4-6节)，花瓣与花丝明显可见的雄蕊形成完成 期(第7-9节)，以及心皮完全愈合且形成分支状 柱头的雌蕊形成完成期(第10～14节)。进一步 研究发现番木瓜花器形成的进程与其所着生的节 数有稳定的一致关系，提示生产上可利用节位标 定的方法来推测芽体的发育程度。还发现番木瓜 花器形成的向顶分化模式和花器形成所需时间会 依季节不同在4--10周之间变动；而花器成熟所 需时间则较为恒定，约为3～5周。在雄蕊形成期 间不同阶段发生的发育受阻现象，则会形成不同 心皮数及柱头数的雌蕊，同时，卢秉国以雌 株、两性株和雄株3种不同类型的番木瓜为材料， 研究了不同类型的番木瓜的花粉育性，观察了大 孢子、雌配子体以及胚胎的发生及发育，研究了育 性对植株结实性能的影响。所有这些研究都在一 定程度上探索了番木瓜不同类型的花器及胚胎发 育过程，使人们对该复杂的过程有了一定认识。 　　 　　4 展望 　　 长期以来，番木瓜主要采用种子进行实生苗 繁殖育苗，但幼苗的雄株率一直维持在5％左右。 由于缺乏有效的性别鉴定方法，只有等到开花时 才能判断，这极大地影响了番木瓜生产，如何在苗 期快速准确地判断、剔除雄株一直是近年研究和 努力解决的问题。形态学鉴定方法简单易行，但 耗时费力；采用生理生化指标鉴别，易受到环境条 件、植株发育阶段和操作人员技术水平等的影响 而导致可靠性降低；快速准确的分子标记辅助选 择技术虽有一定应用，但操作复杂、费用较高，而 且技术的可靠性仍需进一步提高。因此，直到目 前仍缺乏快速、便捷、高效的鉴别手段预测番木瓜 的株型。随着研究和诊断水平的不断提高，性能 稳定、快速简便的鉴定技术必将为番木瓜的早期 高效鉴定提供强有力的判定依据。 相对而言，目前情况下，采用一些廉价的化学 药剂如NAA或GA-3处理番木瓜的幼苗应该是 一条株型早期鉴别的捷径，这种性别控制的方法 操作相当简单，效果较为显著，可以在生产中推广 应用。另一种较为有前途的控制性别的途径是传 统的组织培养法，即对已鉴定为雌株或两性株的 优良品种，进行大规模的组织培养，进而获得大量 遗传同质的雌株或两性株个体供应生产；也可以 对雌株或两性株的花药进行离体培养，获得单倍 体植株后，再用适当浓度的化学药剂，如秋水仙素 或富民隆进行染色体加倍，得到遗传同质的二倍 体雌株或两性株个体，这种组织培养技术的要求 远没有分子鉴定的复杂，设施简单，耗材廉价易 得，而且操作也已基本程序化，许多苗木公司都拥 有自己的组培车间，因此，组织培养法生产遗传同 质的雌株或两性株具有极大的推广应用前景。 研究番木瓜性别遗传及性别决定的规律，在 未来定向改良番木瓜的性别方面具有重要意义， 通过深入研究性别遗传，弄清其遗传规律，并进一 步发现控制性别的基因(如雌性性别的基因)，最
